Every title record that you find in Destiny will have a call number/spine label already, but it needs some editing to match your library’s organization.
As you get to know a library you’ll learn little quirks to their system, but for starters..

You need a title record in Destiny before you can add a copy under that title. The title record contains information like title, author and ISBN. The copy record you add to the title record will contain information like call number and the Destiny barcode.
The book you wish to catalog doesn't show up in any Destiny search results, including a search by ISBN, Title or Author. See here for Destiny Basics.
After searching in under catalog/add title, at the top right corner of the search results will be a link that says Nothing Matches? Add the title
Type in as much information as you think is relevant for the library's purposes. At a minimum you'll want to include title, author and ISBN. When it comes time to add a call number you can search classify by OCLC, http://classify.oclc.org/classify2/ to find out what other libraries have used. You can also find subject headings at classify if you want this book to be found when searching by subjects.
